The holes don't line up with the defender case on, but I would rather fuss with opening the headphone jack cover than taking the case off every time I exercise.
So if you have a protective case and don't want to transition between the both my recommendation is to find a thinner protection case of you want to use this product.

Product seems durable for the price, I like the extra pocket for credit cards or extra cash stitched behind the opening of where you place the phone.

I am not very muscular, but the strap barely was able to grasp the Velcro and almost felt like it was cutting off circulation.
You basically feel like you cut circulation to use this thing, my entire run this felt rather uncomfortable, i had to put it on my forearm (no way this would fit around my bicep)

My phone's touchscreen also works very well even when its inside this arm band so shuffling songs is very easy while running.
